Electrophysiological consequences of vincristine contained chemotherapy in children: A cohort study

Abstract: Vincristine is commonly used in treatment of acute lymphoblastic leukemia (ALL), and peripheral neuropathy is its dose-related toxicity. Motor impairment, specifically muscle weakness is the most severe manifestation of neuropathy. This study aimed at evaluation of the electrophysiological consequences of vincristine-contained chemotherapy in children. In a prospective cohort setting, the electrophysiological studies was performed in 42 children (25 cases of ALL, 17 cases of non-ALL malignancies) at the electr… Show more

“…But contradictory results regarding the relationship between VIPN and age at study have been reported ( Velde et al, 2017 ). Toopchizadeha and others have shown that women have a higher incidence of VIPN ( Barzegar et al, 2015 ), whereas other study found boys to be at higher risk or did not find a significant relation between sex and VIPN in children ( Velde et al, 2017 ). Patients with obesity, diabetes, Charcott-Marie-tooth and obstructive liver disease develop severe VIPN after treatment with VCR ( Gidding et al, 1999 ; Emery et al, 2016 ; Hershman et al, 2016 ; Sajdyk et al, 2020 ).…”

“…While the reported incidence rates vary considerably—depending on the severity of symptoms, diagnostic criteria and concomitant use of other neurotoxic agents—most patients receiving vincristine develop some degree of sensory peripheral neuropathy at a cumulative dose of >4 mg/m 2 (Ramchandren et al, 2009; Toopchizadeh et al, 2009; Seretny et al, 2014; Lavoie Smith et al, 2015; Tay et al, 2017). …”

“…Unfortunately, most patients receiving vincristine develop a dose-dependent peripheral neuropathy characterized by sensory, motoric, and autonomic disturbances (Ramchandren et al, 2009; Toopchizadeh et al, 2009; Seretny et al, 2014; Lavoie Smith et al, 2015). Sensory symptoms occur most frequently, typically develop within several weeks of treatment, and commonly manifest as numbness, loss of sensory discrimination, tingling, and pain (Barton et al, 2011).…”
