Electrospray Ionization and Time-of-Flight Mass Spectrometric Method for Simultaneous Determination of Spermidine and Spermine

Abstract: A sensitive method for the determination of polyamines in mammalian cells was described using electrospray ionization and time-of-flight mass spectrometer. This method was 50-fold more sensitive than the previous method using ionspray ionization and quadrupole mass spectrometer. The method employed the partial purification and derivatization of polyamines, but allowed a measurement of multiple samples which contained picomol amounts of polyamines. Time required for data acquisition of one sample was approximat… Show more

“…However, the derivative procedure was selected since polyamines do not contain a suitable chromophore or fluorophore group and possess the properties of low volatility and low molecular weight. Therefore, derivatization using a suitable labeling reagent is a key step in GC‐MS (Chen et al ., ; Paik et al ., ) and LC‐MS (Byun et al ., ; Liu et al ., ; Moriya et al ., ; Samejima et al ., , ) analyses of polyamines. There is a significant number of precolumn labeling methods using derivatization reagents, for example, isobutyl chloroformate (Byun et al ., ), dansyl chloride (Ducros et al ., ), benzoyl chloride (Liu et al ., ; Asotra et al ., ), N ‐ethoxycarbonyl‐ N ‐pentafluoropropionyl (Paik et al ., ) and N ‐heptafluorobutyryl (Li et al ., ) for the analysis of the polyamines.…”

“…The cell extracts were centrifuged (12 000 g, 10 min, 4 8C), and the supernatant was used for the measurement of putrescine, spermidine and spermine contents. The amount of putrescine, spermidine and spermine was quantitated by mass spectrometric analysis (Samejima et al 2007). …”
