Electrospun Weak Anion-Exchange Fibrous Membranes for Protein Purification

Abstract: Membrane based ion-exchange (IEX) and hydrophobic interaction chromatography (HIC) for protein purification is often used to remove impurities and aggregates operated under the flow-through mode. IEX and HIC are also limited by capacity and recovery when operated under bind-and-elute mode for the fractionation of proteins. Electrospun nanofibrous membrane is characterized by its high surface area to volume ratio and high permeability. Here tertiary amine ligands are grafted onto the electrospun polysulfone (PS… Show more

“…The degree of grafting (DG) yields were estimated in the range of from 0.14% to 2.3% using the following Equation (3). where, W 0 is the weight of unmodified membrane, and W 1 is the weight after grafting the polymer chain [ 42 ]. Polymerization was observed in the membrane surface morphology ( Figure 2 a–f).…”

“…The UV system (Uvaprint 100) was procured from Honle UV America Inc., MA. The SBMA solution and membrane sample were covered by the quartz petri dish and placed into the UV reaction with a UV lamp intensity of 45 mW/cm 2 for different time intervals (PSBMA x ; x: 30 s, 60 s, 120 s, 180 s and 300 s) [ 42 ]. After the modification, the residual chemicals in the solution, monomer SBMA, BP, and poly (SBMA) were washed in DI water and methanol.…”

“…The commercial membrane was soaked in 50% ethanol/DI water for at least 3 h to remove the impurities and glycerin on the membrane surface. The wet membrane sample was rinsed with DI water 3 times, followed by drying in a vacuum oven at 30 °C for at least 12 h. Similar to our previous work, the surface of PVDF ultrafiltration membrane was modified through UV-grafting technique [ 27 , 30 ]. Briefly, the membrane was immersed in 2% BP/methanol solution for 30 min, which was used as the photoinitiator to generate the free-radical on the membrane.…”

“…(2020) using optimized UV‐initiated polymerization, developed a tertiary amine‐functionalized polyacrylonitrile nanofiber membranes that exhibited binding capacities of up to 100 mg mL −1 and recoveries greater than 96%. [ 62 ] Although the above studies are based on model proteins, the outcomes and strategies hold promise and apply to the capture of mAbs. Regenerated cellulose nanofiber membranes functionalized with protein A/G have also been assessed for IgG capture.…”
