Long-term exposure to arsenic has been linked to tumorigenesis in different organs and tissues, such as skin; however, the detailed mechanism remains unclear. In this present study, we integrated "omics" including microRNAome, proteomics and metabolomics to investigate the potential molecular mechanisms. Compared with non-malignant human keratinocytes (HaCaT), twenty-six miRNAs were significantly altered in arsenic-induced transformed cells. Among these miRNAs, the differential expression of six miRNAs was confirmed using Q-RT-PCR, representing potential oxidative stress genes. Two-dimensional gel electrophoresis (2D-PAGE) and mass spectrometry (MS) were performed to identify the differential expression of proteins in arsenic-induced transformed cells, and twelve proteins were significantly changed. Several proteins were associated with oxidative stress and carcinogenesis including heat shock protein beta-1 (HSPB1), peroxiredoxin-2 (PRDX2). Using ultraperformance liquid chromatography and Q-TOF mass spectrometry (UPLC/Q-TOF MS), 68 metabolites including glutathione, fumaric acid, citric acid, phenylalanine, and tyrosine, related to redox metabolism, glutathione metabolism, citrate cycle, met cycle, phenylalanine and tyrosine metabolism were identified and quantified. Taken together, these results indicated that arsenic-induced transformed cells exhibit alterations in miRNA, protein and metabolite profiles providing novel insights into arsenic-induced cell malignant transformation and identifying early potential biomarkers for cutaneous squamous cell carcinoma induced by arsenic.

The industrial augmentation and unguided anthropogenic activities contaminate water sources in most parts of the world especially in developing countries like Pakistan. High concentration of pollutants in groundwater affects human, soil, and crop health badly. The present study was conducted to investigate groundwater quality for drinking and irrigation purposes in an industrial zone of Pakistan. A GIS tool was used to investigate the spatial distribution of different physico-chemical parameters. In this study, the average results exceeding World Health Organization (WHO) and National Environmental Quality Standards (NEQS) were found for pH 7.84, total dissolved solids (TDS) 1492 mg/L, phosphate 0.51 mg/L, dissolved oxygen (DO) 9.92% saturation, F-coli 6.48 colonies/100 mL, Na+ 366 mg/L, HCO3− 771 mg/L, sulfate 251 mg/L, chlorides 427 mg/L, total hardness (as CaCO3) 292 mg/L, electrical conductivity (EC) 2408 μS/cm, iron (Fe) 0.48 mg/L, chrome (Cr) 0.50 mg/L, arsenic (As) 0.04 mg/L, total phosphorus (TP) 0.17 mg/L, sodium adsorption ratio (SAR) 9.76 (in meq/L), residual sodium carbonate (RSC) 9.28 meq/L, % ion balance 14.4 (in meq/L), percentage sodium ion (% Na+) concentration 58.9 meq/L, and water quality index (WQI) 69.0. The trend of cations and anions were (in meq/L) Na > Mg > Ca > K and HCO3 > Cl > CO3 > SO4 respectively. Although the results of the present study showed poor conditions of the groundwater for drinking as WQI but and irrigation purposes as SAR, it needs to improve some more conditions for the provision of safe drinking water and irrigation water quality.

In this study, the quality of groundwater used for drinking purposes at Rajanpur Tehsil Pakistan is investigated. Water contamination causes several health problems to human being and the severity of problems mainly depends upon the level of pollutants present in water. Physical, chemical, and biological parameters of groundwater for twelve locations are analyzed using standard procedures. The obtained results are compared with the standards set by WHO. Most of the chemical parameters are found within the WHO standards except Arsenic, Calcium, and Sulfate at various locations. The Arsenic is found beyond WHO standards at nine locations with an increased percentage of 400% at three locations namely village Mehre Wala, Sehnwal, Near Basti Bhayan, and is also found beyond the permissible limit, i.e., upto 150% at six locations Sikhaniwala, Kot Mithan, Basti Nazru, Basti Asni, Jalal Pur, and Qutab Pul. Only three locations show satisfactory results, namely village shikarpur, village Murghai and Basti Lakha. The percentage of calcium at five locations such as Murghai, Basti Nazru, Basti Asni, Basti Lakha, and Chak Jalal Pur (Pull Pathan) is found to be 44%, 12%, 17.33%, 76%, and 49.33%, which is much higher than the WHO standards. Meanwhile, the Sulphate is also found to be beyond WHO standards at four locations, i.e. 30.8% for Mehrewala, 37.6% for Murghai, 34.8% for Basti Lakha and 34% for Chak Jalal Pur (Pull Pathan). Biological contamination including total coliform and fecal coliform are found to be beyond the limit at Sehanwala, Rangpur, Shikarpur, and Basti Nazru locations. The higher percentage of Arsenic, calcium, and sulfate in water indicate that the groundwater is not suitable for drinking purpose. Sustainable water treatment is highly recommended for drinking water at the identified contaminated location.
