Elicitation effects of oligosaccharides on the transcriptional level of bacitracin ABC transporter genes in Bacillus licheniformis

Abstract: Bacitracin ABC transporter comprises a self-resistance system in Bacillus licheniformis against its own product (bacitracin). Elicitation effects of carbohydrates on transcription level of bcrABC genes encoding bacitracin ABC transporter system were investigated through quantification by real-time PCR. Increases of 68% and 29% in bacitracin A yields after 36 h were achieved in cultures supplemented with oligoguluronate and oligoguluronate/mannan oligosaccharide combination. Cultures with combined elicitors sho… Show more

“…25−27 Regular PCR or qPCR was also run on manure samples for bacitracin resistance genes bceA and bceR 28 as well as bcrA, bcrB, and bcrC. 29 The detection limit for each qPCR protocol was determined as the minimum concentration in the standard curve within the linear range. Key qPCR parameters and the linear range for each primer set can be found in SI Table S3.…”

“…DNA extracts were quantified using a NanoDrop spectrometer. qPCR conditions for tetracycline resistance genes tet (Q) and tet (X) and for tylosin resistance genes erm (B) and erm (F) were adopted from published studies. Regular PCR or qPCR was also run on manure samples for bacitracin resistance genes bceA and bceR as well as bcrA , bcrB , and bcrC . The detection limit for each qPCR protocol was determined as the minimum concentration in the standard curve within the linear range.…”

“…Regular PCR was also run on bacitracin-manure samples for bacitracin resistance genes bceA and bceR (Yoshida et al, 2011) as well as bcrA, bcrB, and bcrC (Murphy et al, 2008). All qPCR results on ARGs were normalized to the 16S rRNA gene, which was quantified using the qPCR protocol from Suzuki et al (2000).…”

“…Evidence for transcriptional level control was obtained using real‐time PCR, where a direct correlation between enhancement of bacitracin A production and the transcription of its biosynthetic genes was made. Elicitor supplementation (OG and MO as above) resulted in increased transcription of the bacitracin biosynthetic genes bacABC ( Murphy et al ., 2007b ), and overexpression of bcrABC genes encoding the bacitracin ABC transporter system ( Murphy et al ., 2008 ). Polysaccharides have also been used to enhance production of secondary metabolites in bacteria and fungi ( Patterson and Bolis, 1997 ; Zhu et al ., 2008 ).…”
