Elucidating the Biosynthetic Pathway for Vibralactone: A Pancreatic Lipase Inhibitor with a Fused Bicyclic β‐Lactone

Abstract: Not so simple: Evidence from 13C‐labeling studies, metabolite profiling, and cell‐free conversion established that the bicycle skeleton of vibralactone is derived from an aryl ring moiety and both shikimate and phenylalanine pathways may contribute. VibPT was found to be potentially involved in vibralactone biosynthesis, as well as an aromatic PTase, which was first characterized in basidiomycete fungi. OPP=pyrophosphate.

“…In recent years, endeavors to explore the potential of producing diverse metabolites of B. vibrans by scale-up fermentation and/or changing culture medium compositions led to the isolation of plenty of bioactive vibralactone congeners and vibralactone biosynthetic-associated compounds [ 3 11 ], including vibralactoximes A–P which represented the first class of natural oxime esters [ 3 ]. The biosynthesis of vibralactone and divergent biosynthetic pathway for skeletally distinct vibralactone-associated compounds were illuminated [ 12 , 13 ]. It was proved that all these compounds shared the biosynthetic precursor 3-prenyl-4-hydroxybenzylalcohol.…”

“…From culture broths of the basidiomycete fungus Boreostereum vibrans (syn. Stereum vibrans ), vibralactones with distinctly skeletons were identified [59] and generated by unusual and divergent biosynthetic pathways that we have recently established by precursor feedings in combination with genome mining [10, 11]. Further analyses of our B. vibrans genome draft assembly revealed several sequences to encode putative sesquiterpene synthases (STS).…”
