Embryonic stem cell markers expression in cancers

Abstract: Abstract:The transcription factors Oct4 and Sox2 are highly expressed in embryonic stem (ES) cells. In conjunction with Klf4 and c-Myc, their over-expression can induce pluripotency in both mouse and human somatic cells, indicating that these factors are key regulators of the signaling network necessary for ES cell pluripotency. Self-renewal is a hallmark of stem cells and cancer and stemness program could play an important role in cancer. Therefore we compared the expression of Oct4, Sox2, Klf4 and c-Myc in 40… Show more

“…This gene is expressed at high levels in neuroepithelial stem cells and its expression persists in neural stem/progenitor cells throughout adulthood (Miyagi et al, 2006). Sox2 is highly expressed in ES and EC cells (Perrett et al, 2008;Schoenhals et al, 2009) and several studies have focused on modulating its expression in these cells in order to elucidate the role of this transcription factor in stem cell pluripotency and self-renewal (Zhao et al, 2004;Boer et al, 2007;Masui et al, 2007;Kopp et al, 2008;Adachi et al, 2010).…”

“…5 Furthermore, after undergoing the differentiation therapy of Yin et al, 1 the gene expression levels of c-Myc and Klf4 became lower than that of Oct3/4 and Sox2. This was comparable to the cases of human iPS cells.…”

“…Notably, KLF4 collaborating with other transcription factors including Oct4, Sox2 and c-Myc, drives somatic cells into iPS cells. CSCs have been identified in various tumors, and KLF4 can be speculated to have similar functions in CSCs based on its function in ES cell [133]. Our work provides evidence for the first time that KLF4 is essential for the maintenance of breast CSC and cell migration and invasion, which may be helpful to develop new therapeutic strategies for breast cancer.…”
