Emerging Function and Clinical Significance of Exosomal circRNAs in Cancer

Abstract: Exosomes are a type of extracellular vesicles (EVs) secreted by almost all cells, with a diameter range of 30-150 nm and a lipid bilayer membrane. Exosomes are now considered as vital mediators of intercellular communication and participate in multiple cellular processes, such as signal transduction and antigen presentation. Recently, circular RNAs (circRNAs), a novel class of noncoding RNAs (ncRNAs), have been found to be abundant and stable in exosomes. Increasing evidence indicates that exosome-derived circ… Show more

“…The expression profile of exosomal circRNA is different from that of producer cells ( Li et al, 2015 ; Dou et al, 2016 ), suggesting that circRNAs are actively incorporated into exosomes. Multiple mechanisms have been implicated in RNA sorting into exosomes, including specific RNA sequences and/or secondary structures associated with RNA binding proteins ( O’Brien et al, 2020 ; Wang M. et al, 2020 ). However, the precise mechanisms through which circRNAs are sorted into exosomes are not yet clear.…”

“…Long non-coding RNAs (lncRNAs) have attracted increasing attention as cancer biomarkers for early screening, diagnosis, prognosis and analysis of treatment responses ( 3 , 4 ). Circular RNA (circRNA) is a new member of the non-coding cancer genome and exhibits distinct properties and diverse cellular functions ( 5 , 6 ). To facilitate the study of cancer-associated lncRNAs, we previously reported the first and second version of the Lnc2Cancer database (Lnc2Cancer 1.0 and 2.0), which enabled users to search all known experimentally supported lncRNAs associated with various human cancers ( 7 , 8 ).…”

“…Exosomes exist in multifarious body fluids, including blood, urine, saliva, bronchoalveolar lavage fluid, follicle fluid, and amniotic fluid. 4 Exosomes are composed of proteins, lipids, glycoconjugates, and nucleic acids. Exosomes contain a number of soluble proteins in the lumen that are found to be associated with the membranes, including integral membrane proteins, lipid-anchored membrane proteins, and peripherally associated membrane proteins.…”
