Enantioselective Demethylation of Nicotine as a Mechanism for Variable Nornicotine Composition in Tobacco Leaf

Abstract: Background: Nornicotine composition in tobacco leaf has been a puzzle for more than half a century. Results: Three recombinant nicotine demethylases selectively use (R)-nicotine in vitro and in planta. Conclusion: Nornicotine composition in tobacco leaf can be reasonably explained by the combination of three nicotine demethylases. Significance: This knowledge lays the base for the optimization of nornicotine composition in tobacco leaf in the future.

“…Nicotine biosynthesis requires an oxidative deamination of N-methylputrescine, catalyzed by N-methyl putrescine oxidase (MPO1) [GenBank: AB289456.1] [17]. The conversion of nicotine to nornicotine involves N -demethylation of nicotine, catalyzed by different isotypes of a cytochrome P 450 enzyme [15], [29], [43]. Two alleles of this gene, CYP82E, were tested: Ntab CYP82E5 v 2 [GenBank: EU182719.1] is found in green leaves of N .…”

“…Nornicotine, 3‐(2‐pyrrolidinyl) pyridine, is a minor alkaloid in tobacco plants. It is the N ‐demethylation product of nicotine which is a predominant tobacco alkaloid and represents 90–95% of the total alkaloid content, and this reaction occurred primarily in tobacco leaves under the mediation of nicotine demethylase enzymes . During the maturation, curing, and processing of tobacco leaves, nornicotine may be N ‐nitrosated to N ′‐nitrosonornicotine (NNN), a well characterized carcinogenic tobacco specific nitrosamines in tobacco products .…”
