Endocervical swabs transported in first void urine as combined specimens in the detection of Mycoplasma genitalium by real-time PCR

Abstract: The aim of this study was to determine whether a patient's endocervical swab specimen can be transported in first void urine (FVU) as combined specimens for the detection of Mycoplasma genitalium by real-time PCR. The study also compared two different DNA extraction methods for observation of possible PCR inhibition. Three specimens, one endocervical swab specimen transported in 2-SP medium, one endocervical swab specimen transported in FVU and a FVU specimen, were collected from 329 women. All sample types un… Show more

“…In women, combination of swabs from two sites results in a better detection rate: 95.7% for combined vaginal and endocervical swabs and 95% when an endocervical swab and FVU specimen are used together. 82,83 Some centres allow patients to obtain their own rectal and vaginal swab specimens because this method of collection has been shown to be as effective as swabs obtained by clinical staff. 8486 Rectal swabs for primarily men-who-have-sex-with-men (MSM) have been utilised.…”

“…an endocervical specimen was taken witb a polyacrylamide swab (Copan, Brescia, Italy), after tbe sampling for C trachomatis, and tbe swab was placed in one of tbe FCU-containing tubes. Tbis combined sample bas been sbown to be accurate in a previous study (16). Tbe specimens were sent to Department of Clinical Microbiology, Linköping University Hospital.…”

“…4,9,13 Theoretically, the combination of vaginal swabs with FCU could increase sensitivity, compared with each specimen alone, as has been demonstrated for the combination of endocervical swabs and FCU in C. trachomatis-infected and Mycoplasma genitalium-infected women. 8,14 There is, however, limited previous knowledge as to whether combined vaginal and FCU specimens yield a similar beneficial outcome. To our knowledge there is only one published study to date, the outcome of which supported the hypothesis.…”
