Endocytosis of Apolipoprotein A-V by Members of the Low Density Lipoprotein Receptor and the Vps10p Domain Receptor Families

Abstract: Apolipoprotein A-V (apoA-V) is present in low amounts in plasma and has been found to modulate triacylglycerol levels in humans and in animal models. ApoA-V displays affinity for members of the low density lipoprotein receptor (LDL-R) gene family, known as the classical lipoprotein receptors, including LRP1 and SorLA/LR11. In addition to LDL-A binding repeats, the mosaic receptor SorLA/LR11 also possesses a Vps10p domain. Here we show that apoA-V also binds to sortilin, a receptor from the Vsp10p domain gene f… Show more

“…Role of SorLA in Regulating GDNF Secretion-Considerable evidence supports that SorLA has multifunctional roles in Golgi-endosomal transport (21,24,33). The interaction of SorLA with GDNF, the localization of SorLA to GDNF-containing vesicles, and the assemblage in Golgi of both proteins prompts the hypothesis that SorLA might regulate the secretion of GDNF.…”

“…Although the exact mechanism underlying the effects of sortilin on lipid metabolism has not been fully examined, sortilin binds to LDL on the cell surface 30) and increases the amount of LDL internalized 18,31) . In addition, sortilin has been shown to bind apolipoprotein A-V and lipoprotein lipase 32,33) . The effect of sortilin on very low-density lipoprotein (VLDL) synthesis, as determined by overexpression and knockdown studies, is conflicting 11,34,35) .…”

“…Meanwhile, additional analyses will be required to exclude causal roles for other genes (i.e., CELSR2 and PSRC1) that map closer to the strongest associations across a neighboring recombination hotspot to SORT1. Recently, SORT1 has been connected to the endocytosis of ApoA-V-containing chylomicrons (Nilsson et al, 2008). The main SNP near SORT1 gene is associated with LDL-C and ApoB, and this association is most significant in the whole genome scan.…”
