Endophytes Aspergillus caespitosus LK12 and Phoma sp. LK13 of Moringa peregrina produce gibberellins and improve rice plant growth

Abstract: Two new strains of endophytic fungi were isolated from the bark of Moringa peregrina and identified as Aspergillus caespitosus LK12 and Phoma sp. LK13. These endophytes were identified through amplifying polymerase chain reaction (PCR) and sequencing the 18S internal transcribed spacer of DNA extracted from both endophytes. Pure cultures of endophytic fungi were subjected to extract and isolate gibberellins (GAs). H 2 ]-GA 7 were used to quantify the endophytic fungal GAs. The analysis revealed that both the e… Show more

“…have characterized GAs using capillary GC/MS. GA analysis using HPLC coupled with GC/MS-SIM gives more reliable results than those obtained using thin-layer chromatography, bioassays, or HPLC with ultraviolet detection, which have poor resolution and decreased reliability Waqas et al 2013;Khan et al 2014). GC/MS-SIM is an established method for identifying targeted novel secondary metabolites, and our results were confirmed when we detected no GAs in the bacterialfree culture broth treated as same using negative control.…”

“…They are associated with numerous plant species and isolated from nearly all organs of every plant sampled (Currie et al 2014). Endophytes enhance plant growth by the production of plant hormones such as indole-3-acetic acid (IAA), gibberellins (GAs) and cytokinins (Khan et al 2014). Antimicrobial metabolites like colletotric acid (Zou et al 2000), griseofulvin (Park et al 2005) and glucosidase inhibitor 3S, 4R-(?…”

“…Gibberellins were also found in supernatants of endophytic Aspergillus caespitosus and Phoma spp. (Khan et al, 2014) and P. citrinum (Khan et al, 2008). It should be highlighted that this funding is the first report showing the growthpromoting ability of the most bioactive isolates P. crustosum (I74) and F. proliferatum (I92) recovered from S. linnaeanum.…”
