Endophytic Fungi from Lycium chinense Mill and Characterization of Two New Korean Records of Colletotrichum

Abstract: Chinese boxthorn or matrimony vine (Lycium chinense Mill) is found primarily in southeastern Europe and Asia, including Korea. The dried ripe fruits are commonly used as oriental medicinal purposes. Endophytic fungi were isolated from surface sterilized tissues and fruits of the medicinal plant in 2013 to identify the new or unreported species in Korea. Among 14 isolates, 10 morphospecies were selected for molecular identification with the internal transcribed spacer (ITS) gene. Phylogenetic analysis revealed … Show more

“…TS2′s ITS1/ITS2/D1-D2 concatenated sequence-based phylogeny was obtained by BioNJ using K2P as a substitution model, both of which have been previously used to estimate fungal phylogenetic relationships from ribosomal markers [72,73]. K2P has also been applied to study taxonomic divergence in different taxa such as bacteria and protozoa [74,75].…”

“…The species was also found, as A. sclerotigenum , to be a year-round endophyte of Quercus ilex in north-central Spain [ 33 ]. Barcode type 1 isolates, listed as Acremonium sp., were obtained in Korea as an endophyte of the medicinal Chinese boxwood, Lycium chinense [ 34 ]. A. egyptiacum was a moderately prevalent isolate from diseased or dead larvae of the rose stem girdler beetle Agrilus aurichalcenus in the Kerman Province of south-eastern Iran [ 35 ].…”

“…Varieties inoculated with pathogens were Juhwangmi, Dahomi, Yulmi, Pungwonmi, Sinjami, and Jinhongmi. Three storage roots from each variety were washed in running tap water and sterilized as previously described methods [ 27 ] with minor modifications. Samples were then washed three times with distilled water to remove surface sterilizing agents.…”
