Endopolyploidy in Vanda Miss Joaquim (Orchidaceae)

Abstract: Summary• Occurrence of endopolyploidy in somatic tissues of the hybrid orchid Vanda Miss Joaquim ( Vanda hookeriana × Vanda teres ) was investigated with respect to tissue type and developmental stage. Effects of naphthaleneacetic acid (NAA) and gibberellic acid (GA 3 ) on endopolyploidy during embryo development were also studied.• For the study of endopolyploidy, flow cytometric analysis was employed to determine nuclear DNA content of cells of somatic tissues.• Multiploid cells were observed in leaves, root… Show more

“…The enlargement of the nucleus and the elongation of the whole cell to a final size that is larger than that of the surrounding cells that are still dividing are the first observed changes in developing crystal idioblasts (Franceschi and Horner, 1980;Kausch and Horner, 1983a). For raphide crystal idioblasts of V. planifolia (Orchidaceae) and of Vanda, a close relative of Phalaenopsis, this enlargement is coupled with endopolyploidization (Kausch and Horner, 1984;Lim and Loh, 2003), which is caused by a switch of their cell cycle program from growth to endopolyploidization and cell enlargement (Sugimoto-Shirasu and Roberts, 2003;Jakoby and Schnittger, 2004). The observation that HSS is not expressed in these specialized cells suggests that HSS expression and most probably PA biosynthesis are restricted to meristematic (i.e.…”

“…Systemic somatic endopolyploidization has been reported in many plant species (de Rocher et al, 1990;Galbraith et al, 1991;Gilissen et al, 1993;Kudo and Kimura, 2001;Barow and Meister, 2003;Lim and Loh, 2003). Those studies suggest very frequent occurrence of endopolyploidy in angiosperms, but it is characteristic for certain families.…”
