Endothelin-1 as a Local Ovarian Regulator in Porcine Granulosa Cells

Abstract: This study was conducted to investigate whether endothelin-1 (ET-1) has direct effects on the functions of porcine granulosa cells harvested from small or medium follicles. Positive ET-1-like immunoreactivity (ET-l-LI) was observed in the cultured porcine granulosa cells, and Northern blot analysis demonstrated the expression of mRNA for prepro-ET-1 in these cells. The binding study showed the presence of a nonselective, single class of binding sites which predominantly included the subtype ETB. Inc… Show more

“…ET-1 produced by these cells may function as an autocrine/paracrine growth factor and modulator of steroidogenesis [Kamada et al, 1993]. Positive ET-1 immunoreactivity and Northern blot expression of mRNA for prepro-ET-1 in cultured porcine granulosa cells also suggest that these substances exert autocrine/paracrine effects on the function of granulosa cells and that ET-1 acts as modulator of steroid metabolism in preovulatory follicles [Kubota et al, 1994]. Autoradiographic studies have shown that in the pig granulosa cells contain ET receptors and that they are also distributed in the vascular components of the ovary [Flores et al, 1995].…”

“…ET-1 could then act through ET B receptors on the capillary endothelial cells of the corpora lutea and through ET A receptors on the vascular smooth muscle cells in the media of stromal vessels. An autocrine action on granulosa cells was suggested by the presence of ET receptors (Kamada et al 1995(Kamada et al , 1992Kubota et al 1994;Tedeschi et al 1994), the synthesis of ET-1 (Iwai et al 1991;Kamada et al 1993a;Magini et al 1996) and ET-1 mRNAs (Kubota et al 1994;Magini et al 1996), and the expression of ECE-1.…”

“…Cultured porcine granulosa cell media contain ET-1 (Iwai et al 1991;Kamada et al 1993a). Cultured porcine granulosa cells express ET-1 mRNA (Kubota et al 1994). Human granulosa cells in situ would contain ET-1 peptide and mRNAs at different stages of maturation (Magini et al 1996).…”

“…In contrast, there have been several reports on the physiological roles of ET-1 in uterus and ovary. Expression of the ET-1 gene was detected in human and porcine granulosa cells (Kubota et al 1994, Magini et al 1996. The peptide is considered to play an important role in granulosa cell function.…”
