Ferroptosis is a nonapoptotic form of cell death characterized by iron-dependent accumulation of lipid hydroperoxides to lethal levels. Necroptosis, an alternative form of programmed necrosis, is regulated by receptor-interacting protein (RIP) 1 activation and by RIP3 and mixed-lineage kinase domain-like (MLKL) phosphorylation. Ferroptosis and necroptosis both play important roles in the pathological progress in ischemic stroke, which is a complex brain disease regulated by several cell death pathways. In the past few years, increasing evidence has suggested that the crosstalk occurs between necroptosis and ferroptosis in ischemic stroke. However, the potential links between ferroptosis and necroptosis in ischemic stroke have not been elucidated yet. Hence, in this review, we overview and analyze the mechanism underlying the crosstalk between necroptosis and ferroptosis in ischemic stroke. And we find that iron overload, one mechanism of ferroptosis, leads to mitochondrial permeability transition pore (MPTP) opening, which aggravates RIP1 phosphorylation and contributes to necroptosis. In addition, heat shock protein 90 (HSP90) induces necroptosis and ferroptosis by promoting RIP1 phosphorylation and suppressing glutathione peroxidase 4 (GPX4) activation. In this work, we try to deliver a new perspective in the exploration of novel therapeutic targets for the treatment of ischemic stroke.

The consumption of cycad (Cycas circinalis) seeds has been linked to the development of Amyotrophic Lateral Sclerosis-Parkinsonism Dementia Complex (ALS-PDC) in humans. ALS-PDC is a clinically variable disease presenting as a combination of symptoms typical of PD and/or ALS. Chronic consumption of β-sitosterol β-D-glucoside (BSSG), a component of the cycad seed, by rats (Rattus norvegicus) has been previously reported to initiate a progressive pathology that develops over several months and manifests as behavioural and histopathological changes that resemble characteristic features of Parkinson’s disease. As part of an independent multi-site validation study, we have tried to replicate and further characterize the BSSG model with a focus on motor function, and associated immunohistochemical markers. Beginning at 3 months of age, male CD® (Sprague Dawley) rats (N = 80) were dosed orally with either a flour pellet or a flour pellet containing BSSG (3 mg) daily (5×/week) for 16 weeks consistent with previous reports of the model. Following BSSG intoxication, separate cohorts of animals (n = 10/treatment) were exposed to a behavioural test battery at 16, 24, 32, or 40 weeks post-initial BSSG feeding. The test battery consisted of the open field test, cylinder test, and ultrasonic vocalization (USV) assessment. No changes in behaviour were observed at any time point. Following behavioural testing, animals were processed for immunohistochemical markers of substantia nigra integrity. Immunohistochemistry of brain tissue revealed no differences in the microglial marker, Iba1, or the dopaminergic integrity marker, tyrosine hydroxylase (TH), in the substantia nigra at any assessment point. The absence of any group differences in behaviour and immunhistochemistry indicates an inability to replicate previous reports. Further investigation into the sources of variability in the model is necessary prior to further utilization of the BSSG model in preclinical studies.

Elevated visceral adiposity index is associated with increased stroke prevalence and earlier age at first stroke onset: Based on a national cross-sectional study

ObjectiveThe purpose of this study was to examine the association between the VAI (visceral adiposity index) and stroke prevalence and age at stroke in US adults.MethodsWe examined the association between VAI and stroke prevalence and age at stroke using logistic regression, subgroup analysis, and dose-response curves using participants from the National Health and Nutrition Examination Survey (NHANES) database from 2007-2018.ResultsThis study ultimately included 29,337 participants aged >20 years, of whom 1022 self-reported a history of stroke, and after adjusting for all confounders, each unit increase in corrected VAI was associated with a 12% increase in the prevalence of stroke (OR= 1.12, 95% CI: 1.01, 1.24) along with an earlier age at stroke 1.64 years (β= -1.64, 95% CI: -2.84, -0.45), stratified analysis showed that the prevalence of stroke was 20% higher in the female group (OR= 1.20, 95% CI: 1.04, 1.39), black group (OR= 1.22, 95% CI: 1.01, 1.48), age ≤60 years group (OR= 1.25, 95% CI: 1.05, 1.48), hypertensive group (OR=1.15, 95% CI:1.01, 1.31), and diabetic group (OR=1.23, 95% CI:1.02, 1.48) VAI increase was positively correlated with stroke prevalence increase. The dose-response curves showed a positive linear correlation between increased VAI and stroke prevalence, while a negative linear correlation was observed between increased VAI and age at stroke.ConclusionAlthough a causal relationship cannot be proven, higher VAI was positively associated with stroke prevalence and can lead to earlier stroke onset.
