Endotoxin Testing

Abstract: SUMMARYParenterals, sterile preparations intended to be injected in man or animal, should be free from pyrogenic substances which are able to raise the thermostatic setting in the hypothalamus. This, article gives an up-to-date review of the principal detection and quantification methods for these agents, with special attention on the chromogenic Limulus Amebocyte Lysate assay.

“…1 Various structural components of bacteria, yeasts, and molds have been isolated and characterized as pyrogenic. 2 Organic and inorganic dusts, nanoparticles, and diesel exhaust particles may also induce an inflammatory reaction, although the nature of their pyrogenic components is not yet known.…”

“…11 The assay can only be performed with liquid samples, thus again only a rinsing solution of the biomaterial can be assessed. 1 The rinsing solution is prepared with purified water, with or without a detergent (Tween 20). By detecting LPS, the LAL detects the most common and most potent pyrogen known with high sensitivity.…”

“…The common Limulus amoebocyte lysate (LAL)-based methods of endotoxin testing are not applicable to endotoxin determination in HBOCs, unless the assay is modified to account for the presence of endotoxin-binding hemoglobin [94][95][96]. Chromogenic method of endotoxin testing is based on the generation of free p-nitroaniline (pNA) by cleavage action of procoagulant enzymes of LAL on a peptide-pNA conjugate and spectrophotometry at 405 nm.…”
