Energy Deposition Events Produced by Fission Neutrons in Aqueous Solutions of Plasmid DNA

Abstract: Using an agarose gel electrophoresis assay, single-strand breaks (ssb) induced by fission neutrons and 60Co gamma-rays in aerobic aqueous solutions of pBR322 plasmid DNA were studied. The energy-deposition events of the two radiations were characterized using a Rossi-type proportional counter to measure lineal-energy spectra. For neutrons, the dose-weighted lineal-energy mean, yD, is 63 keV micron-1--about 30 times that for gamma-rays. With increasing yD, hydroxyl radicals produced within spurs or tracks are l… Show more

“…Afterwards, the gels were scanned by UV light to excite DNA-enhanced EB fluorescence using a Shimadzu CS-9000 thinlayer chromatography scanner coupled to a microcomputer. Most of the previous similar studies applied UV light near 315 nm to excite DNA-EB fluorescence [6,[24][25][26], but here 260 nm was chosen so that the sensitivity in detecting DNA was enhanced by about 4.7 times (unpublished result). The emitted fluorescence of 590 nm was determined through a 550 nm cut-off filter.…”

“…In the experiment conducted under aerobic conditions, the SSB yields caused by γ -rays are predominantly due to OH radicals for scavenging capacities that are cell-like or less than cell-like [38]. The calculated ratio of the OH reaction, 90%, can be explained in terms of the high activity of OH and the oxygen-scavenging effect of H and eaq (Eqs.…”

“…The supercoiled form has less ethidium bromide binding ability than the open circular and linear conformations. Therefore, a correction factor of 1.7 for supercoiled pBR322 has been reported to approximate this difference among the three plasmid conformations [15]. The numbers of SSBs and DSBs were calculated by the following equations [16]:…”
