Energy substrate requirements for in-vitro development of hamster 1- and 2-cell embryos to the blastocyst stage

Abstract: Energy substrate requirements (pyruvate, lactate and amino acids) were determined for in-vitro development of hamster 1- and 2-cell embryos to blastocysts, using a chemically defined, protein free medium (hamster embryo culture medium, HECM). One-cell embryos were very sensitive to energy substrate type and concentration. Pyruvate alone could not support development of 1-cell embryos to greater than 4-cells, whereas lactate as sole energy substrate supported 14% development into morulae/blastocysts. Pyruvate, … Show more

“…The superiority of CZB to WM as a medium for mastomys embryo culture may be attributed to the presence of glutamine. Similar beneficial effects of glutamine on embryonic development were reported in hamster [15], pig [16], sheep [17], and a certain blocking mouse strain [7]. Glutamine is known to be used by preimplantation mammalian embryos as sources of energy or amino groups [18].…”

“…Chemically defined media have been developed for IVM and IVC in rodents and domestic animals (McKiernan et al, 1991;Keskintepe and Brackett, 1996;McKiernan and Bavister, 2000;Kishida et al, 2004;Oyamada et al, 2004). Recent work reported the success of two chemically defined media (CMRL-1066 and HECM-10) in supporting maturation of rhesus monkey oocytes in vitro (Zheng et al, 2001a).…”

“…Energy substrates have been found to affect oocyte maturation and embryo development in a wide variety of mammals (McKiernan et al, 1991;Rose-Hellekant et al, 1998;Zheng et al, 2001c). Glucose, lactate and pyruvate are designated as the "energy substrate triad" in cellular metabolism (Bavister, 1995).…”

“…In order to control the variance derived from individual animals, the COCs from individual females were equally assigned to all the treatment groups in experiments 2 and 3 (controlled pooling) [16]. Percentage data were transformed by arcsin transformation (Tukey-Freeman transformation) [17], and nuclear numbers, by log transformation.…”
