Engagement of the Mannose Receptor by Tumoral Mucins Activates an Immune Suppressive Phenotype in Human Tumor‐Associated Macrophages

Abstract: Tumor-Associated Macrophages (TAMs) are abundantly present in the stroma of solid tumors and modulate several important biological processes, such as neoangiogenesis, cancer cell proliferation and invasion, and suppression of adaptive immune responses. Myeloid C-type lectin receptors (CLRs) constitute a large family of transmembrane carbohydrate-binding receptors that recognize pathogens as well as endogenous glycoproteins. Several lines of evidence demonstrate that some CLRs can inhibit the immune response. I… Show more

“…Ligation of MMR-1 is also linked to immune regulation. In some reports, activation of MMR-1 on human moDCs resulted in IL-10 secretion and reduced T cell stimulatory activity of DCs (40,41). As expected from these publications, we found a significant increase in IL-10 production of cells that were stimulated with AG alone.…”

“…Such T-cell irresponsiveness has been shown to inhibit the development of experimental autoimmune encephalomyelitis after injection of mannosylated myelin peptide (4). Second, MR engagement on monocyte-derived DCs induces an immune-suppressive phenotype (32,33). Third, substantial evidence points out that MR expression is directly correlated with the immunogenicity of the cellular environment.…”

“…34,35 Both TNF-α and IκBα siRNA decrease mRNA for CD206 in macrophages ( Figure 8C). The combination of the two treatments also significantly decreases CD206 mRNA.…”

“…33 We have functionalized the surface of these particles with mannose as a targeting ligand in order to facilitate uptake of the particles in TAMs, which have increased amounts of mannose receptor (CD206) on their surface. 34,35 The mannose receptor is an endosomal pattern-recognition receptor, which, when present at high levels, is associated with suppression of the classical Figure 1 Overview of the two arms of the NF-κB pathway. Notes: The classical pathway is broadly activated by immunostimulatory molecules: TNF-α, lPs, Il-1, IFNγ, and others.…”
