Engineering of Hydroxymandelate Oxidase and Cascade Reactions for High-Yielding Conversion of Racemic Mandelic Acids to Phenylglyoxylic Acids and (R)- and (S)-Phenylglycines

Abstract: An alcohol oxidase (AOx) for the (S)-enantioselective oxidation of 4-hydroxymandelic acid (4-HMA) to 4-hydroxyphenylglyoxylic acid (4-HPGA) with enhanced activity was developed by directed evolution of hydroxymandelate oxidase (HMO) through three rounds of iterative saturation mutagenesis. The engineered HMO mutant A80G-T159S-T162Q (HMOTM) catalyzed the oxidation of (S)-4-HMA to 4-HPGA with a 23-fold enhancement in catalytic efficiency (k cat/K M). Substrate docking simulation on HMOTM suggested that A80G reor… Show more

“…The reduction amination catalyzed by GluDH was thermodynamically favorable with a high equilibrium constant (Keq = 10 14 –10 18 ) (Cheng et al 2020 ), which ensured the complete conversion of α-KG to L-Glu. The cascade that coupled GluDH and transaminase had great potential for unnatural amino acid biosynthesis, and several successful applications were developed, such as 1,2-amino alcohols and ( R )- and ( S )-phenylglycines (Jung et al 2023 ; Liu et al 2019 ). By employing this cascade system, 400 mM PPO was completely converted to L-PPT using only 40 mM amino donor L-Glu.…”

“…For these, additional extensive HMO engineering is required. [30] A four-enzyme cascade from styrene to (R)-phenylethanolamine (R-PEA) and a six-enzyme cascade from L-phenylalanine to (R)-PEA were developed. In both cascades, the alditol oxidase (AldO) catalysed the second last reaction in converting the intermediate (R)-1-phenyl-1,2-ethanediol (R-PED) to (R)-mandelic aldehyde (MALD).…”

“…In particular, derivatives with halogen substituents in para ‐position suffered from poor conversion and were not included in the final cascade. For these, additional extensive HMO engineering is required [30] …”
