Fast and highly sensitive detection has become critical to prevent the spread of severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2). However, fast analysis tends to reduce the sensitivity and accuracy of such method. This study is to investigate a fast and highly sensitive screening method for detecting polymerase chain reaction (PCR) products of SARS‐CoV‐2 through voltage‐programmed capillary gel electrophoretic separation. By separating and detecting the PCR products of RNA‐dependent RNA polymerase (RdRp) and spike (S) genes, we have diagnosed coronavirus disease 2019 (COVID‐19) cases within 4 min using such method with high reliability and accuracy, indicating at least a 7.5‐fold faster than previous COVID‐19 diagnostic methods. The detection limits of RdRp and S genes fragments were 1.34–3.46 pM. The developed method can be used to analyze PCR products with specific DNA sizes as well as detect SARS‐CoV‐2 mutations without losing resolution. Furthermore, it can also be used for the fast analysis of DNA molecules related with various infectious diseases.

Alzheimer’s disease (AD) is an irreversible neurodegenerative disease with clinical symptoms of memory loss and cognitive impairment. Currently, no effective drug or therapeutic method is available for curing this disease. The major strategy used is to identify and block AD at its initial stage. Thus, early diagnosis is very important for intervention of the disease and assessment of drug efficacy. The gold standards of clinical diagnosis include the measurement of AD biomarkers in cerebrospinal fluid and positron emission tomography imaging of the brain for amyloid-β (Aβ) deposits. However, these methods are difficult to apply to the general screening of a large aging population because of their high cost, radioactivity and inaccessibility. Comparatively, blood sample detection is less invasive and more accessible for the diagnosis of AD. Hence, a variety of assays based on fluorescence analysis, surface-enhanced Raman scattering, electrochemistry, etc., were developed for the detection of AD biomarkers in blood. These methods play significant roles in recognizing asymptomatic AD and predicting the course of the disease. In a clinical setting, the combination of blood biomarker detection with brain imaging may enhance the accuracy of early diagnosis. Fluorescence-sensing techniques can be used not only to detect the levels of biomarkers in blood but also to image biomarkers in the brain in real time due to their low toxicity, high sensitivity and good biocompatibility. In this review, we summarize the newly developed fluorescent sensing platforms and their application in detecting and imaging biomarkers of AD, such as Aβ and tau in the last five years, and discuss their prospects for clinical applications.

This work proposes controllable molecular sieving using the cross-linked copoly(poly(ethylene glycol) acrylate/poly(ethylene glycol) diacrylate) (copoly(PEGA/PEGDA)) hydrogels as a sieving matrix for DNA in capillary electrophoresis (CE). Attempts were made to control the separation performance using different gel components, including the initiator concentration (C APS), total concentration (C t) of monomers, the ratio (R) of PEGA to PEGDA, and the length of the cross-linker (L c). Results reveal that the molecular sieving effect in the copoly(PEGA/PEGDA) gel can be controlled by C t, R, C APS, and L c, especially by the former two parameters. Higher C t and lower R provided smaller average pore size, resulting in better separation of small-size DNA fragments, while lower C t and higher R allowed better separation of large-size DNA fragments. Moreover, using a representing gel component, the repeated test showed a run-to-run precision of RSD < 0.9% (n = 10 tests), a column-to-column precision of RSD < 5.9% (n = 5 columns), and a day-to-day precision of RSD < 5.2% (n = 25 tests in 5 days) for the migration time. The results also reveal the rapid and high-resolution separation for 20–140 bp DNA fragments using the copoly(PEGA/PEGDA) gel when tuned to a lower R and lower C t. When high C t was used, the copoly(PEGA/PEGDA) gel capillary was found to have better repeatability and higher resolution than the authentic poly(acrylamide) gels owing to the better volume stability upon polymerization.
