Enhanced Cisplatin Chemotherapy by Iron Oxide Nanocarrier-Mediated Generation of Highly Toxic Reactive Oxygen Species

Abstract: Reactive oxygen species (ROS) plays a key role in therapeutic effects as well as side effects of platinum drugs. Cisplatin mediates activation of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX), which triggers oxygen (O) to superoxide radical (O) and its downstream HO. Through the Fenton's reaction, HO could be catalyzed by Fe/Fe to the toxic hydroxyl radicals (OH), which cause oxidative damages to lipids, proteins, and DNA. By taking the full advantage of Fenton's chemistry, we herein demons… Show more

“…A more direct method is to develop iron‐containing nanoparticles that can deliver and release iron into cancer cell. Most previous studies used solid iron‐based nanocrystals, such as iron oxide nanoparticles and FePt nanoparticles as the iron source to trigger Fenton chemistry in tumor . The amount of Fe 2+ /Fe 3+ ions released from these nanoparticles is influenced by pH value and reductive environment .…”

“…In addition, unlike cisplatin, the nanoparticles are internalized into cells via endocytosis, which contributes to the reversal of the cell’s resistance to cisplatin. [47] …”

“…Ma et al first used cisplatin to activate NOX for catalyzing dissolved O 2 into O 2 •− , which can be further converted into H 2 O 2 by cellular SOD (Equation ), finally favoring IONP‐triggered Fenton reaction . Based on the intrinsic catalytic chemistry of glucose oxidase (GOx), which can catalyze the production of H 2 O 2 from intratumoral glucose (Equation ), we recently constructed GOx and Fe 3 O 4 nanoparticles coloaded dendritic MSNs (DMSNs) for improved CDT efficacy ( Figure ) .…”
