Enhanced dispersive solid phase extraction assisted by cloud point strategy prior to fluorometric determination of anti-hepatitis C drug velpatasvir in pharmaceutical tablets and body fluids

Abstract: Highly sensitive and selective solid phase extraction method combined with cloud point extraction strategy was developed for analysis of velpatasvir in its pharmaceutical tablets and biological fluids.

“…The obtained data indicated the high precision of the proposed method at the repeatability and intermediate levels as discovered by the small values of RSD (Table ). Also, accuracy and precision were further examined through statistical analysis of the results obtained by the proposed and reported spectrofluorimetric method using Student's t ‐test and variance ratio F ‐test. Results showed no significant differences between the two methods regarding accuracy and precision (Table ).…”

“…Only one fluorometric and few liquid chromatographic methods were reported for determination of VLP . To the best of our knowledge, until now, no article has been issued providing stability indicating assays using the spectrofluorimetric technique for the estimation of VLP.…”

“…Although the provided literature reference was very limited, remarkable sensitivity and good accuracy were obtained when applied to real samples, as well as good sorbent reusability, attesting to the high potential of the proposed methods and sorbents ( Table 4 ). An interesting method proposed by El-Wekil et al saw the use of cobalt hydroxide nanoparticles (CHNPs) as sorbents complementary to reduced graphene oxide (rGO) in the dSPE procedure [ 44 ]. CHNP addition inhibited graphene aggregation and therefore ensured the high surface area of the sorbent, as well as decreasing its binding affinity towards the surfactant, polyethylene glycol (PEG) 6000, used in a cloud point extraction (CPE) system, via the formation of rGO-CHNPs hydrogen bonds.…”

“…Pure VLP showed a pH dependent release, where it showed good dissolution up to pH of 2, after which it decreased drastically as shown in Figure 7. This could possibly be due to the low solubility of VLP at higher pH values [40,41,[69][70][71].…”
