Enhanced production of acetoin and butanediol in recombinant Enterobacter aerogenes carrying Vitreoscilla hemoglobin gene

Abstract: Microbial production of butanediol and acetoin has received increasing interest because of their diverse potential practical uses. Although both products are fermentative in nature, their optimal production requires a low level of oxygen. In this study, the use of a recombinant oxygen uptake system on production of these metabolites was investigated. Enterobacter aerogenes was transformed with a pUC8-based plasmid carrying the gene (vgb) encoding Vitreoscilla (bacterial)hemoglobin (VHb). The presence of vgb an… Show more

“…The compound 2,3-butanedione was the most important precursors of alkylpyrazine (28,29). The compound 3-hydroxy-2-butanone, which is the immediate precursor to 2,3-butanediol, can be oxidized to 2,3-butanedione for use as a natural flavour in dairy products (30,31). Furfural and benzaldehyde were also detected in most of the Zhenjiang aromatic vinegars, which were responsible for empyreumatic, caramel and a pleasant almond-like odour.…”

“…Converti and Perego (2002) conducted metabolic flux analyses with various initial glucose concentration conditions. Geckil et al (2004) tried to increase 2,3-butanediol production by introducing the hemoglobin gene (vgb) of Vitreoscilla.…”

“…Metabolic changes in other bacteria engineered to express VHb have also been reported [Geckil et al, 2004;Liu et al, 1995;Wei et al, 1998]. The most systematic survey of metabolic effects related to VHb has been done in E. coli using microarrays to assess changes in gene expression at the RNA level [Roos et al, 2004].…”
