Enhanced upregulation of smooth muscle related transcripts by TGF 2 in asthmatic (myo) fibroblasts

Abstract: Background: Transforming growth factor beta (TGFb) upregulates a number of smooth muscle specific genes in (myo)fibroblasts. As asthma is characterised by an increase in airway smooth muscle, we postulated that TGFb 2 favours differentiation of asthmatic (myo)fibroblasts towards a smooth muscle phenotype. Methods: Primary fibroblasts were grown from bronchial biopsy specimens from normal (n = 6) and asthmatic (n = 7) donors and treated with TGFb 2 to induce myofibroblast differentiation. The most stable genes … Show more

“…Myofibroblast differentiation was assessed using the common myofibroblast marker, a-SMA, by immunocytochemistry, Western blot, Imagestream flow cytometry, and RT-PCR. In addition, the expression of a novel myofibroblast (37). CDDO, 15 d-PGJ 2 , and rosiglitazone all inhibited myofibroblast differentiation, as determined by Western blot for a-SMA ( Figure 1A) and by immunocytochemistry ( Figure E1).…”

“…These data have led us to suggest that PPARg agonists are a promising intervention to prevent tobacco smoke-induced asthma in offspring (10,11). It is important to point out that the mesenchymal markers of airway reactivity chosen by us are markers of myofibroblast differentiation (a-SMA and calponin), a hallmark of chronic lung diseases, such as asthma, and/or have been shown to have increased matrix deposition (fibronectin and collagen III) in the subepithelial space of the airways in asthma (18)(19)(20). Furthermore, the other markers used by us, such as b-catenin and LEF-1, are functional intermediates in the Wnt signaling pathway, a key mediator of myogenesis, justifying their inclusion in our analysis (21)(22)(23).…”

“…RNA was extracted using TRIzol reagent (Invitrogen), DNase treated, and reverse-transcribed for quantitative PCR (qPCR) as previously described (16). Genes of interest were normalized to the geometric means of ubiquitin C and phospholipase A2.…”
