Enhancement of ambulation-increasing effect of methamphetamine by peripherally-administered 6R-L-erythro-5,6,7,8-tetrahydrobiopterin (R-THBP) in mice.

Abstract: Abstract-Behavioral effects of 6R-L-erythro-5,6,7,8-tetra hydrobiopterin (R THBP), a co-factor for tyrosine hydroxylase and tryptophan hydroxylase, were investigated by means of ambulatory activity in mice. Single administration of R THBP (50 and 100 mg/kg, s.c.) showed no significant effect on the mouse's ambulatory activity for 5 hr. The ambulation-increasing effect of methamphetamine (2 mg/kg, s.c.) was dramatically enhanced and prolonged by the pretreatment with R-THBP (100 mg/kg, s.c.) 0, 2, 6, 12 and 24 … Show more

“…However, it is interesting that R-THBP did not en hance the ambulatory increment by ketamine. R-THBP was reported to markedly augment the effect of methamphetamine, which was intimately connected with the newly synthesized dopamine (34). It is prob able that ketamine stimulates the dopamine release, but hardly affects the dopamine synthesis, unlike metham phetamine.…”

“…Imipramine and R THBP enhanced the ambulation-increasing effect of methamphetamine, cocaine and/or GBR-12909 even though the tested doses of these 3 drugs were subeffective or effective (11,19). Furtheremore, 0.1 mg/kg of MK-801, which was a subeffective dose for increasing the mouse's ambulatory activity, did not show any interaction with the drugs mentioned above.…”

“…On the other hand, R-THBP enhanced the ambulation-increasing effect of GBR, simi lar to methamphetamine. We have demon strated that R-THBP produces no significant change in the effect of cocaine (21). Further more, the modifications by various drugs of the ambulation-increasing effect of GBR sug gest that GBR may interact, probably indirect ly, with noradrenergic, cholinergic and adeno sine systems, etc.…”

“…GBR was effective for enhancing the ambulation-increasing effects of all these drugs. R-THBP acts as a coenzyme for tyrosine hydroxylase (20), and it markedly enhances the ambulation-increasing effect of methamphetamine (21). This com pound also enhanced the effect of GBR.…”
