This study aimed to determine the effect of enrofloxacin (ENR) on the transfer of the plasmid-mediated quinolone resistance (PMQR) gene qnrS from opportunistic pathogen Escherichia coli (E2) to Salmonella Enteritidis (SE211) and to analyze the resistance characteristics of SE211-qnrS isolates. The plasmid carrying qnrS gene of E2 was sequenced by Oxford Nanopore technology. The plasmid carrying qnrS gene belonged to incompatibility group IncY. In vitro, the transfer experiment of IncY plasmid was performed by the liquid medium conjugation method. The conjugation transfer frequency of the IncY plasmid was 0.008 ± 0.0006 in the absence of ENR, 0.012 ± 0.003 in 1/32 MICENR, 0.01 ± 0.008 in 1/8 MICENR, and 0.03 ± 0.015 (Mean±SD) in 1/2 MICENR, respectively. After inoculation of E. coli E2 and SE211, chickens were treated with different doses of ENR (3.03, 10, and 50 mg/kg b.w.) for 7 days consecutively. To screen the SE211-qnrS strains from intestinal tract of chickens, the resistance genes and susceptibility of isolates were identified. The amount of E. coli E2 and the copy number of qnrS gene in the chicken intestinal tract were determined by colony counting and qPCR, respectively. In vivo, more SE211-qnrS strains were isolated from the treated group compared with the untreated group. SE211-qnrS strains not only obtained IncY plasmid, but also showed similar resistance phenotype as E2. In conclusion, ENR treatment can promote the spread of a IncY-resistance plasmid carrying the qnrS fluoroquinolone-resistance gene in Escherichia coli and the development of drug-resistant bacteria.

During the past few decades, organochlorine pesticides (OCPs) have emerged as global pollutants. Despite their ban, Endosulfan, one of the most persistent and toxic pesticides of the OCP group, was most commonly used in agriculture and other sectors. Endosulfan bioremediation experiments were conducted using Pleurotus ostreatus. The Endosulfan spiked wheat straw was inoculated with Pleurotus ostreatus and incubated under solid state and submerged fermentation conditions. The enzyme production and activities and degradation of Endosulfan isomers (α-and β-isomers) was monitored periodically for 40 days. Degradation rate and half-life (DT50) of both the isomers was calculated using a simple first-order kinetics model. Under both conditions, the addition of Endosulfan showed stimulating effects on MnP and laccase enzyme activities. Activities of both the enzymes at each time interval were higher in solidstate fermentation than submerged fermentation. The degradation of α-isomers was higher under solidstate fermentation than submerged fermentation, while the inverse was true for β-isomer. The calculated DT 50 of α-and β-Endosulfan under solid-state fermentation was 3.99 and 44 days, respectively, with 73.69±3.43 mg kg-1 Endosulfan sulfate accumulation. While under submerged fermentation conditions, DT 50 of α-and β-Endosulfan was 10 and 20 days, respectively. The formation of Endosulfan sulfate, under submerged fermentation conditions, was maximum at day 10 (39.67±5.73 mgL-1) and declined to 8.71±3.24 mgL-1 until the end of incubation. A poor correlation between degradation and activities of both enzymes was observed. Rapid reduction in Endosulfan sulfate under submerged fermentation revealed its ascendancy in degradation over solid-state fermentation.

The practice of incorporating animal manure into soil is supported within the European Circular economy as a possible substitute for mineral fertilizers and will become crucial for the sustainability of agriculture. However, this practice may indirectly contribute to the dissemination of antibiotics, resistance bacteria, and resistance genes. In this study, medicated drinking water and poultry litter samples were obtained from a broiler-chick farm. The obtained poultry litter was incorporated into the soil at the experimental field site. The objectives of this research project were first to develop analytical methods able to quantify fluoroquinolones (FQs) in medicated drinking water, poultry litter, and soil samples by LC–MS; second to study the fate of these FQs in the soil environment after incorporation of poultry litter from flock medicated by enrofloxacin (ENR); and third to screen the occurrence of selected fluoroquinolone resistance encoding genes in poultry litter and soil samples (PCR analysis). FQs were quantified in the broiler farm’s medicated drinking water (41.0 ± 0.3 mg∙L−1 of ENR) and poultry litter (up to 70 mg∙kg−1 of FQs). The persistence of FQs in the soil environment over 112 days was monitored and evaluated (ENR concentrations ranged from 36 μg∙kg−1 to 9 μg∙kg−1 after 100 days). The presence of resistance genes was confirmed in both poultry litter and soil samples, in agreement with the risk assessment for the selection of AMR in soil based on ENR concentrations. This work provides a new, comprehensive perspective on the entry and long-term fate of antimicrobials in the terrestrial environment and their consequences after the incorporation of poultry litter into agricultural fields.
