Envelope glycoproteins from human immunodeficiency virus types 1 and 2 and simian immunodeficiency virus can use human CCR5 as a coreceptor for viral entry and make direct CD4-dependent interactions with this chemokine receptor

Abstract: Several members of the chemokine receptor family have recently been identified as coreceptors, with CD4, for entry of human immunodeficiency virus type 1 (HIV-1) into target cells. In this report, we show that the envelope glycoproteins of several strains of HIV-2 and simian immunodeficiency virus (SIV) employ the same chemokine receptors for infection. Envelope glycoproteins from HIV-2 use CCR5 or CXCR4, while those from several strains of SIV use CCR5. Our data indicate also that some viral envelopes can use… Show more

“…The CCR5 chemokine coreceptor appears to be one of the most significant structures in mediating binding and infection by macrophage (M)-tropic HIV in humans [1,2] and both M-and T-tropic simian immunodeficiency virus (SIV) in macaques [3][4][5]. The importance of CCR5 in HIV infection has been amply demonstrated by the homozygous ¿ 32 deletion of CCR5 in about 1 % of caucasian populations [6][7][8].…”

“…While the global loss of CD4+ lymphocytes can be caused by the virus-mediated destruction of the infected cells, a selective depletion of some CD4 + cells may occur as a result of the increased susceptibility of these cells to the virus infection. Among those factors underlying the susceptible depletion of CD4 + lymphocytes may be the activation status [8,10,16,[18][19][20][21][22] and selective expression of chemokine receptors in the CD4+ lymphocytes [6, 11,171. On the other 174 hand, antigen stimulation during the persistent virus infection may continuously drive the clonal activation and expansion [ 121.…”

“…As follows from recent experimental data [14,15], it seems that besides CD4 and CCR5, there are other coreceptors promoting M-tropic virus entry into cells. To date, the information is scarce, although some such receptors have been described recently [10,15±17].…”

“…A number of experimental observations have suggested a greater range and complexity of coreceptors for the immunodeficiency viruses than previously described [14,15]. In fact, some new virus cofactors have recently been discovered.…”
