Epidermal growth factor as a diuretic in sheep.

Abstract: 1. Epidermal growth factor (EGF) has recently been shown to stimulate both polydipsia and polyuria and the aim of this study was to determine which was the primary response. Ewes received a continuous intravenous saline infusion (100 ml day1) for 12 days (days 1-12) and EGF at doses of 0 (n = 6) or 10 jug h' (n = 6) over days 5-8. The supply of water was ad libitum during days 1-4 and 9-12, but was fixed at the pretreatment mean of days 1-4 for each ewe during days 5-8. 2. During the period of fixed water inta… Show more

“…16 The idea that EGF may play a role in water balance was previously suggested by Gow and Phillips, 17 because they showed that EGF infusion in sheep resulted in brisk diuresis and natriuresis. Therefore, we used erlotinib, a Food and Drug Administration (FDA) -approved EGFR inhibitor, 18,19 to study the underlying pathways affected by EGF inhibition in water balance.…”

“…Where a significant interaction was obtained, simple main effects were determined. The a priori data of interest were those comparing EGF treatment versus control treatment, EGF + indomethacin treatment versus indomethacin treatment and EGF + indomethacin treatment versus EGF treatment during the period of treatment (days 9-12) and the subsequent period after this (days [13][14][15][16][17][18][19][20]. Statistical differences between individual means were calculated using the Newman-Keul's post hoc comparisons test.…”

“…The pharmacological interest in the peptide for use in humans has mainly focused on its role in accelerating wound healing and tissue repair by local and systemic applications [7]. Recently, it was also demonstrated that administration of EGF had an isolated diuretic effect in rats [8], and it has been suggested that EGF antagonizes vasopressin action inducing diuresis in sheep [9, 10]. However, the cellular mechanism for this is not completely understood.…”
