Epigenetics in Liver Fibrosis

Abstract: Liver fibrosis is a common consequence of chronic liver injury and is a key determinant of liver-associated morbidity and mortality. Identification of new mechanisms of fibrosis, including disease-specific molecular drivers, remains relevant to reveal novel biomarkers and therapeutic targets. Recently, greater accessibility to more advanced molecular methods that can assess changes in epigenetic regulation has stimulated more research investigating the epigenetic landscape of liver fibrosis. Such studies have … Show more

“…Unfortunately, HDACIs have been shown to carry side effects such as nausea, vomiting, fever, and headache, which can be easily managed [ 139 ]. However, serious adverse effects, such as hepatotoxicity, have also been observed [ 140 ], including a clinical case of steatohepatitis and hepatocellular carcinoma [ 141 ]. VPA has been associated with dose-dependent elevation of serum aminotransferases [ 142 ].…”

“…The distinct anatomical location and proposed role of environmental factors such as excessive alcohol consumption (13), heavy manual labor, and exposure to vibrations (14) suggest epigenetic regulation is likely to be important in the pathogenesis of the disease. Epigenetic mechanisms have been shown to be crucial mediators of fibrotic gene expression in a wide range of fibrotic disorders (15), including cardiac fibrosis (16), liver fibrosis (17), and systemic sclerosis (18), and probably account for the persistence of myofibroblasts in disease (19).…”

“…Activated HSCs are thought to be major mediators of liver fibrosis, and their generation is maintained during liver fibrogenesis [5]. The epigenetic machinery, including DNA methylation, plays a crucial role in the transition of quiescent HSCs to activated HSCs [6][7][8]. It is expected that epigenetic therapy will be effective in changing the phenotype of HSCs from activated to quiescent and in inhibiting liver fibrogenesis [9,10].…”
