Epigenome-wide association study in healthy individuals identifies significant associations with DNA methylation and PBMC extract VEGF-A concentration

Abstract: Introduction: Vascular endothelial growth factor A (VEGF-A) is a chemokine that induces proliferation and migration of vascular endothelial cells and is essential for both physiological and pathological angiogenesis. It is known for its high heritability (> 60%) and involvement in most common morbidities, which makes it a potentially interesting biomarker. Large GWAS studies have already assessed polymorphisms related to VEGF-A. However, no previous research has provided epigenome-wide insight in regulation of… Show more

“…However, epigenetic modifications may diminish VEGF expression. Vesna and colleagues found twenty significant CpG sites in promoter regions, suggesting that the concentration of VEGF-A can be regulated by DNA methylation ( 77 ). Based on The Cancer Genome Atlas (TCGA) databases, Zhou and colleagues discovered that overexpression of insulin-like growth factor 2 mRNA-binding protein 3 (IGF2BP3) was related to cancer progression and poor survival of patients.…”

“…Interestingly, 20 methylation sites have been significantly associated with VEGFA levels in PBMCs from healthy individuals. 38 Therefore, methylation levels at these sites could be evaluated in affected patients, in order to identify potential alterations that could serve as therapeutical target to influence VEGFA expression and activity. Furthermore, AMD-related epigenetic marks and events characterizing molecular factors involved in Angiopoietin/Tie-2 pathway could be investigated.…”

“…The accumulation of high-throughput genome-wide methylation data has enabled the analysis of human methylomes across distinct populations and medical cohorts via epigenome-wide association studies (EWAS) ( Gorenjak et al , 2020 ; Küpers et al , 2019 ; Chu et al , 2017 ). Such analyses have shown that predisposition to common human disease is frequently associated with specific methylation signatures in distal control regions also known as gene enhancers ( Li et al , 2013 ).…”
