Epitope analysis of Ara h 2 and Ara h 6: characteristic patterns of IgE‐binding fingerprints among individuals with similar clinical histories

Abstract: Summary Background Ara h 2 and Ara h 6 are moderately homologous and highly potent peanut allergens. Objective To identify IgE-binding linear epitopes of Ara h 6, compare them to those of Ara h 2, and to stratify binding based on clinical histories. Methods Thirty highly peanut-allergic subjects were stratified by clinical history. Sera were diluted to contain the same amount of anti-peanut IgE. IgE binding to overlapping 20-mer peptides of Ara h 2 and Ara h 6 was assessed using microarrays. Results Eac… Show more

“…Known linear epitopes of Ara h 2 and 6 recognized by allergic patients are highlighted in colour . Of the 3 isoforms, Ara h 7.0201 showed most sequence similarity with Ara h 2.0201 and 6.01 in the C‐terminal regions that are known to be allergenic linear epitopes in Ara h 2 and 6 (orange underlined sequence) . Similar to the conserved cysteine pattern of at least 8 conglutins of Ara h 2.0201 and Ara h 6.01, Ara h 7.0201 is the only isoform containing 8 cysteine residues (underlined C‐residues), whereas Ara h 7.0101 and Ara h 7.0301 only contain 6 cysteine residues.…”

“…In addition to the sequence alignment, predictive protein 3D models of all isoforms were therefore created with PHYRE and UCSF Chimera (Figure ). Known allergenic linear epitopes of Ara h 2.0201 and 6.01 are highlighted in the same colours as displayed in Figure (Figure A, B) . Figure C‐E shows the predicted 3D models of the Ara h 7 isoforms.…”

“…202 Ara h 6 shares 55% identity with the AA sequence of Ara h 2 and some of the IgE epitopes of Ara h 6 are cross-reactive with those of Ara h 2. 173 As with Ara h 2, the resistance of Ara h 6 to heat and digestive enzymes was reported. 201 Seven IgE-binding epitopes were identified for Ara h 6 by Otsu et al, 173 and the linear IgEbinding epitopes of Ara h 2 and Ara h 6 were useful in predicting clinical reactions in patients with peanut allergy.…”

“…200,201 Seven to ten IgE-binding epitopes were identified in Ara h 2 using overlapping peptide immunoassays. 12,39,173,174 In addition, posttranslational proline hydroxylation of Ara h 2 contributes to linear IgE epitopes. 202 Ara h 6 shares 55% identity with the AA sequence of Ara h 2 and some of the IgE epitopes of Ara h 6 are cross-reactive with those of Ara h 2.…”
