EPR Spectroscopic Studies of Lipoxygenases

Abstract: Polyunsaturated fatty acids are sources of diverse natural, and chemically designed products. The enzyme lipoxygenase selectively oxidizes fatty acid acyl chains using controlled free radical chemistry; the products are regio‐ and stereo‐chemically unique hydroperoxides. A conserved structural fold of ≈600 amino acids harbors a long and narrow substrate channel and a well‐shielded catalytic iron. Oxygen, a co‐substrate, is blocked from the active site until a hydrogen atom is abstracted from substrate bis‐ally… Show more

“…[5][6][7] An enzymatically active diamagnetic ion in the biomolecules is replaced with a paramagnetic ion, or diamagnetic molecules are labeled with a paramagnetic ion to study them by electron paramagnetic resonance (EPR) spectroscopy. [8][9][10][11][12][13][14][15] This provides scope for structural characterization of biomolecules [16][17][18][19][20] and understanding the local environment around the paramagnetic center. [21][22][23][24] In view of this, many biologically important metal ions, such as copper(II), [25][26][27] cobalt (II), [28][29][30][31] iron(III), [32][33][34] manganese(II), [35,36] and other ions, [37][38][39] have been studied by EPR spectroscopy.…”

“…Hamadou et al (2020) studied the influence of physicochemical characteristics of neem seeds in the biodiesel production. Gaffney (2020) studied via EPR spectroscopy the lipoxygenases, an important and abundant component in soybean seeds. Chakraborty and Bhattacharjee (2020) investigated substances in mustard seeds via EPR spectroscopy.…”

“…2,6,10−12 The most widely accepted mechanism involves two states for the iron site: a high-spin ferric hydroxide and a high-spin ferrous aqua complex (Scheme 1a). 2,13 In the initial step of the reaction cycle, the ferric hydroxide converts the pentadiene unit into a pentadienyl radical via an overall H atom abstraction (Scheme 1b).…”

“…All novel compounds (1)(2)(3)(4)6) were structurally characterized by X-ray structure analyses. Representative perspective views of the complex cations 1, 2, and 3 are given in Figure 1, including the applied numbering of those atoms relevant to the subsequent discussion.…”
