Equilibration and freezing interactions affecting bull sperm characteristics after thawing

Abstract: ABSTRACT:The objective was to determine effects of equilibration length and freezing curve type as well as their interactions on motility and live spermatozoa proportion in bull sperm after thawing. The ejaculates of 6 sires were repeatedly collected. Fresh semen was diluted with one extender and divided into 3 groups equilibrated for 30, 120, and 240 min. Subsequently, half straws of each group were frozen using standard 3-phase or 2-phase freezing curve differing in the rate of temperature decrease. Th e spe… Show more

“…The factors of bull's individuality or thawing method were detected as important in presented study as well as in many previous (Beran et al, 2013;Paldusová et al, 2016;Doležalová et al, 2016). Both these criteria should be therefore tested and proven in specified conditions to optimize methods of IDs processing.…”

“…Our results monitored immediately after thawing are ambiguous in this connection, when both too slow thawing method as well as rapid thawing method reached better spermatozoa motility than control method of thawing. The individuality of bull and also the semen processing influenced the results of spermatozoa motility after thawing (Robbins et al, 1976;Moore et al, 2006;Beran et al, 2011;Doležalová et al, 2016). Therefore, these factors should serve as possible explanation of various results presented in this study with those of opposite.…”

“…The amount of annually produced cryopreserved ID exceeds several hundred millions, whereas production of fresh doses reaches only one-fifth of that number (Gordon, 2017). A standard preservation procedure consists of several different steps, namely, semen collection, evaluation, dilution, cooling, equilibration and freezing-thawing process in case of cryopreservation (Doležalová et al, 2016;Khalil et al, 2018). Both aforementioned ID production methods provide their own advantages and disadvantages.…”

“…Semen cooling rate induces irreversible changes in membrane composition and functionality (Watson 2000). Other factors affecting the sperm motility and sperm membrane integrity are time of equilibration, freezing rate, and interactions between equilibration time and freezing rate (Dolezalova et al 2016). Cryopreservation process also affects acrosomal integrity and capacitation (Pons-Rejraji et al 2009).…”
