Essential Involvement of Neutrophil Elastase in Acute Acetaminophen Hepatotoxicity Using BALB/c Mice

Abstract: Intense neutrophil infiltration into the liver is a characteristic of acetaminophen-induced acute liver injury. Neutrophil elastase is released by neutrophils during inflammation. To elucidate the involvement of neutrophil elastase in acetaminophen-induced liver injury, we investigated the efficacy of a potent and specific neutrophil elastase inhibitor, sivelestat, in mice with acetaminophen-induced acute liver injury. Intraperitoneal administration of 750 mg/kg of acetaminophen caused severe liver damage, suc… Show more

“…Real-time RT-PCR was performed as described previously 79 , 80 . Briefly, total RNA was extracted from whole kidney samples using ISOGENE (Nippon Gene, Toyama, Japan) according to the manufacturer’s instructions, and 1 μg of total RNA was reverse-transcribed into cDNA at 42 °C for 1 h in 20 μl of a reaction mixture containing mouse Moloney leukemia virus reverse transcriptase (PrimeScript, Takara Bio Inc., Otsu, Japan) with 6 random primers (Takara Bio Inc.).…”

“…NLRP3, a key component of the inflammasome, plays a critical role in the secretion of IL-1β and in pyroptosis, which is an inherently inflammatory caspase-1-dependent mechanism of cell death triggered by various pathological stimuli that have been critically implicated in the pathogenesis of acute lung injury ( Peiró et al, 2017 ; Hsu et al, 2022 ). NE is an active protease released from neutrophils involved in tissue damage by inducing direct cytotoxicity and proinflammatory mediator release ( Ishida et al, 2023 ). Numerous evidence suggests that NE acts as a key target in ALI, and inhibitors of this protease are able to inhibit ALI in patients with cardiopulmonary bypass as well as other animal models associated with ALI ( Pan et al, 2023 ).…”
