Essential Role of STAT3 Signaling in Hair Follicle Homeostasis

Abstract: Dominant-negative mutations associated with signal transducer and activator of transcription 3 (STAT3) signaling, which controls epithelial proliferation in various tissues, lead to atopic dermatitis in hyper IgE syndrome. This dermatitis is thought to be attributed to defects in STAT3 signaling in type 17 helper T cell specification. However, the role of STAT3 signaling in skin epithelial cells remains unclear. We found that STAT3 signaling in keratinocytes is required to maintain skin homeostasis by negative… Show more

“…However, the specific targeting of STAT3 has been challenging. This is particularly critical since STAT3 signaling is also essential in hair follicle and bone homeostasis [ 24 , 25 ]. siRNAs, used for the specific down-regulation of targeted genes, have garnered considerable interest as an attractive new class of drugs for a broad range of clinical applications [ 26 ].…”

“…STAT-3 mutation determined in mice has been indicative of telogen to anagen phase obstruction; pointing that STAT-3 expression in basal keratinocytes through anti-EGFR treatment may give strong evidence of hypertrichosis onset. 14,19 Promising basic research information implicating EGFR, FGF, and STAT-3 with a majority of skin cutaneous hair morphogenesis and multistep transformation giving strong evidence of possible interactive mechanisms of undesirable documented events. So publishing distinct cases combined with EGFR manipulation undoubtedly will focus on a challenging era hinting uninvestigated drug-induced mechanisms.…”

“…STAT3 (Signal Transducer and Activator of Transcription 3) is activated by cytokines and growth factors. This gene plays an important role in maintaining the homeosis of skin (48). STAT5A (Signal Transducer and Activator of Transcription 5A) protein serves a dual function of signal transduction and activation of transcription in cells exposed to cytokine and other growth factors.…”
