Establishing the chromatographic fingerprint of traditional Chinese medicine standard decoction based on quality by design approach: A case study of Licorice

Abstract: A novel analytical quality by design approach for developing a chromatographic fingerprint was established for analyzing complex traditional Chinese medicine, using a licorice standard decoction as an example. Considering the characteristics of integrity and ambiguity, the resolution of eight common peaks, total peak number, capacity factor distributions, and peak purity were selected as potential critical method attributes for assessing the quality of the chromatographic fingerprint. A central composite desig… Show more

“…First, under different elution gradients ( Figures 4A–D ), the retention times and separation degrees of characteristic peaks were optimized to obtain a better chromatogram that could be used in SES-TCMCF ( Figure 4F ). In addition, there were six characteristic peaks ( Figure 4E ) whose correlation coefficients were greater than 0.99 between the integral area and content of the samples ( Liang et al, 2010 ), and they were selected for calculations of the ZNC content ( Jiao et al, 2015 ; Zhang et al, 2019 ). A validation experiment was performed in triplicate tests under the optimized UAE conditions.…”

“…Therefore, in recent years, TCM analysis has begun to focus on the rapid, integrated and comprehensive analysis. A number of methods, such as fingerprints, 38,39 chromatography, 40,41 spectroscopy, 42,43 and multiple component analysis, have been used to control the quality of TCM. Certain biological chromatography approaches based on the absorption, distribution, metabolism and elimination of the drugs have also been used in the analysis of TCM and screening of the bioactive markers.…”
