Establishment of a ciliated epithelial cell line from human Fallopian tube

Abstract: Human tubal epithelial cells in primary culture were transfected with simian virus 40 (SV40) large T antigen plasmid, and an immortalized ciliated cell line, named as NT/T-S, was established without crisis. Transmission electron microscopy proved that NT/T-S cells had cilia, microvilli, junctional complexes, rough endoplasmic reticula, free ribosomes and microtubules. NT/T-S cells were evaluated preliminarily on the basis of co-culture study using surplus embryos at the 4- to 8-cell stage in our IVF and embryo… Show more

“…Previous efforts to recapitulate primary human FTE in culture have produced suboptimal results due to the rapid dedifferentiation and loss of the polarized morphology and the biological behavior of the two sub-populations of epithelium (Ando et al , 2000; Comer et al , 1998; Henriksen et al , 1990; Kervancioglu et al , 1994). …”

“…Propagation of human immortalized ovarian surface cells (IOSE) 84 and fallopian tube fimbria has been made possible by stable transfection with SV40 T antigen. 85,86 For example, IOSE-29 does not form tumors, while its neoplastic counterpart IOSE29EC forms s.c. and i.p. tumors in SCID mice.…”

“…This becomes critical when small animals like mice are used for experiments. Oviductal epithelial cell lines are established from pathological samples of pig and human oviduct (Boullant et al, 1973;Ando et al, 2000;Lee et al, 2001). To our knowledge, no clonal fibloblastic cell lines are yet established from oviducts, and analysis of epithelial-stromal interaction is very important in epithelial differentiation.…”
