Establishment of a New Extrahepatic Bile Duct Carcinoma Cell Line, TFK-1.

Abstract: A new human extrahepatic bile duct carcinoma cell line (TFK-1) was established from a surgically resected tumor specimen, which was histologically diagnosed as partly papillary adenocarcinoma and partly differentiated tubular adenocarcinoma. The tumor cells cultured in RPMI-1640 medium supplemented with 10% FBS grew as monolayers showing epithelial-like morphology with a population doubling time of 37 hr during exponential growth at passage 40. Chromosome number was distributed in the range of 72 to 76, with a… Show more

“…However, cell lines originated from biliary tract cancers have rarely been reported. Ten human extrahepatic bile duct carcinoma cell lines (SK-ChA-1 (Knuth et al, 1985), KMBC (Yano et al, 1992), and OCUCh-LM1 (Yamada et al, 1995), TFK-1 (Saijyo et al, 1995), ICBD-1 (Takiyama et al, 1998), HBDC (Jiao et al, 2000), SCK, JCK, Cho-CK, Choi-CK (Kim et al, 2001a)) and three gall bladder carcinoma cell lines (MZ-ChA-1 and MzChA-2 (Knuth et al, 1985), OCUG-1 (Yamada et al, 1997) have been reported in the world literature.…”

“…The human cholangiocarcinoma cell lines KMCH-1, 19 TFK-1, 20 Mz-ChA-1, 21 and KMBC 22 were cultured in Dulbecco's modified Eagle medium supplemented with 10% fetal bovine serum, penicillin G (100,000 U/L), streptomycin (100 mg/L), and gentamycin (100 mg/L). 11,23 H69 cells, immortalized human cholangiocytes, were kindly provided by Dr. D. Jefferson (Tufts University, Boston, MA) and cultured as previously described.…”

“…Human hepatobiliary carcinoma cell lines used in this study were HuCC-T1, 49 derived from moderately differentiated adenocarcinoma from intrahepatic bile duct carcinoma (IHBC); HuH-28, 50 undifferentiated adenocarcinoma of IHBC; TFK-1, 51 partly papillary adenocarcinoma and partly differentiated tubular adenocarcinoma of extrahepatic bile duct carcinoma (EHBC); NCCC-1, well-differentiated adenocarcinoma of EHBC; and MzChA-1, 52 a highly differentiated papillary adenocarcinoma of GBC.…”
