Establishment of an R-banded rabbit karyotype nomenclature by FISH localization of 23 chromosome-specific genes on both G- and R-banded chromosomes

Abstract: Direct detection of fluorescent in situ hybridization signals on R-banded chromosomes stained with propidium iodide is a rapid and efficient method for constructing cytogenetic maps for species with R-banded standard karyotypes. In this paper, our aim is to establish an R-banded rabbit karyotype nomenclature that is in total agreement with the 1981 G-banded standard nomenclature. For this purpose, we have produced new GTG- and RBG-banded mid-metaphase karyotypes and an updated version of ideograms of R-banded … Show more

“…All rabbit bacterial artificial chromosomes (BACs) were isolated from a rabbit library maintained at the UR1313, Unité de Génétique Animale et Biologie Intégrative, Institut National de la Recherché Agronomique (INRA). Hprt1 and Jarid1c BACs had been previously published 35,36 . Xist-containing BACs (573D5, 614B8 and 717B2) were isolated by a PCR-based screening protocol (C. Rogel-Gaillard, unpublished results).…”

“…Our data on the rabbit SRY suggests that this sex-determining gene is probably duplicated and evolving under concerted evolution in Lagomorphs. The recent identification of a clone that contains this gene [8] in a rabbit BAC library [13] will hopefully allow its complete sequencing in our laboratories and the examination of the hypotheses described above.…”

“…To this end, we performed a FISH analysis using 2 BAC clones as specific probes, either for the rabbit X chromosome (530F8 at Xq26.1) or for the rabbit Y chromosome (828D7 at Yp12) [27]. BAC 530F8 was labeled with Spectrum Green, and BAC 828D7 with Spectrum Red (all from Vysis) by a standard nick translation reaction.…”
