Establishment of functional human pituitary tumor cell cultures

Abstract: Five primary human pituitary tumor cell cultures were initiated from adenoma fragments obtained from patients with prolactin-secreting adenomas and acromegaly. Functional cell cultures were maintained and propagated in monolayer or suspension culture for up to 9 months. Optimal cell viability and growth were achieved using Ham's F10 medium enriched with 20% fetal bovine serum, although cells from a patient with acromegaly also grew in serum-free, defined, hormone-containing medium. Bromocriptine (100 ng/ml) di… Show more

“…Four immortalized cell lines originating from human pituitary adenomas exist to date: HPA [ 49 ], GX [ 50 ], HP75 [ 51 ] and PDFS [ 52 ]. Human primary cultures can be maintained as 2D cultures in vitro for a few passages and are mostly used to decipher the mechanisms of hormone secretion [ 53 , 54 , 55 , 56 ]. Although PitNET organoid literature is sparse, a few publications have emerged in the last few years.…”

“…However, this culture of secretory human pituitary cell tumors is not an efficient development; dispersed tumor pituitary cells grow poorly in culture, the cells exhibit limited attachment and spreading, and the cells typically form floating aggregates in the culture dish 9,10 . Several attempts have been made to obtain a tumor pituitary cell suspension, including an enzymatic and mechanical dispersion approach 11 , a solely mechanical dispersion approach 12 and the culturing of tumor explants 10 . With these approaches, different authors have obtained viable adherent cultures for different periods of time.…”
