Establishment of Glucocorticoid-Mediated Transcriptional Induction of the Rice XA21 Pattern Recognition Receptor

Abstract: The rice pattern recognition receptor, XA21, confers robust resistance to the bacterial pathogen Xanthomonas oryzae pv. oryzae (Xoo). We have generated transgenic plants that express Xa21 only in the presence of the glucocorticoid hormone, dexamethasone (DEX). DEX-mediated transcriptional induction of Xa21 is accompanied by upregulation of pathogenesis-related 1 gene expression and restriction of Xoo multiplication. The DEXinducible system can be used to synchronize the XA21-mediated response to infection thro… Show more

“…However, due to the fact that in many cases non-pathogenic microbes might also possess such molecules, the strict term "pathogen-associated" is limiting and will be replaced in the following by the term MAMP. MAMPs are perceived by corresponding plant pattern recognition receptors (PRR) 23 at low concentrations [24][25][26][27] and display many diverse structures, such as carbohydrates, peptidoglycans, peptides, proteins, lipids, and sterols. [26][27][28][29] MAMPs can be constituents of the outer layers of fungal pathogens, such as chitin (1) fragments, 30,31 of oomycete (Phytophthora) pathogens in the case of b-(1,3)-(1,6)-glucan (2) fragments 32 and Pep-13 (3), a peptide of the cell wall transglutaminase of Phytophthora.…”

“…The growth perturbations were caused probably by the GVG activator that was binding to cis-regulatory elements in the plant genome with sequence homology to GAL4 upon Dex activation (Kang et al, 1999). Nevertheless, the system was used successfully in a number of studies in rice (Chern et al, 2013;Nakayama et al, 2013;Onodera et al, 2018, Park et al, 2012Qu et al, 2009) in which the negative effects were reduced by shortening the time of Dex exposure or selecting lines with low GVG expression levels and mild phenotype that could be used as activator lines.…”

“…In our system, about half of the independently transformed rice lines displayed weak or moderate leaky expression (Park et al , 2012). Therefore, it is critical to identify the transgenic lines that display the least leaky expression while maintaining highly inducible expression.…”

“…To confirm non-leaky expression of Xa21 , selected lines should be fully susceptible to Xoo in the absence of DEX and resistant to Xoo after DEX is applied. Additionally, Xa21 gene expression and protein abundance should be low or not detectable by quantitative RT-PCR and western blot analysis before DEX treatment and should be induced by DEX application (Park et al , 2012). …”
