Establishment of High and Low Metastasis Cell Lines Derived from a Human Tongue Squamous Cell Carcinoma

Abstract: Malignant tumors are composed of cells with different phenotypic properties and only certain cell subpopulations present metastatic potential. The establishment of cell lines with high or low metastatic potential is necessary to investigate the molecular mechanisms of the metastatic process. However, human oral squamous cell carcinoma (SCC) cell lines that are suitable for the above investigation are scarce. High and low metastatic cells were obtained from a primary lesion of a patient with tongue carcinoma wh… Show more

“…17 Decreased levels of E-cadherin have also been associated with a metastatic phenotype in SCCHN cell lines. 18 This relationship has also been shown in tumor specimens, 19 indicating that loss of E-cadherin may be an important step in the development of metastasis in SCCHN.…”

“…In this research, we used the UM1 and UM2 cells. These cells are paired cell lines from a single lingual squamous cell carcinoma patient with different metastatic potentials (13). Specifically, UM1 is more aggressive than UM2 in terms of cell invasion (14).…”

“…The UM1 and UM2 cells used in this study are paired cell lines with different metastatic potential that were generated from a single patient with lingual squamous cell carcinoma (13). These cell lines were maintained in Dulbecco's modified Eagle's medium/F12 (DMEM/F12) supplemented with 10% FBS, 100 U/ml penicillin and 100 µg/ ml streptomycin.…”

“…Human oral cancer cell line, UM1 [52] and the zinc-inducible OAZ transfectant (pMT/CB6 + HuFSAZ-wt) and the mock vector transfectant (UM1-pMT/CB6 + ) were cultured as previously described [25]. The expression of OAZ gene from the pMT/CB6 + HuFSAZ-wt was induced by 100 µM ZnSO 4 treatment and the protein samples were harvested after one week ZnSO 4 treatment.…”
