“…Photolysis of penic Paper chromatography of the hydrazone solution using a propanol-2-water-ammonia solvent (20:2: 1) revealed, after treatment with alkali, two coloured zones, one almost in the solvent front and brown in colour dur to the unreacted hydrazine and the other (RF 0.60) green-brown in colour. Reduction of the hydrazone by zinc and hydrochloric acid as described by Alfthan and Virtanen (1955) and subsequent electrophoresis revealed a ninhydrin-positive spot corresponding to that of penicillamine disulphide, consistent with a keto acid derived from the complete substrate molecule, having the structure HOOC-CO-C(CH3)2-S-S-Pen.…”

“…System 2 was also used for the separation of ribose and 5(4)-amino-4(5)-imidazolecarboxamide from the hydrolysis products of the riboside. In certain experiments, the keto acids isolated from the ether-soluble metabolie products in the medium at the end of the culture period were separated by direct chromatography in solvent system 13 (Wieland & Fischer, 1949;Magasanik & Umbarger, 1950); in others, separation was achieved by the method of Alfthan & Virtanen (1955), the amino acids obtained on reduction of the 2:4-dinitrophenylhydrazones being identified by chromatography in solvent system 2 in parallel with authentic amino acids as reference compounds. Solvent system 6 was used in conjunction with solvent system 2 for two-dimensional chromatography of protein hydrolysates according to the method of Levy & Chung (1953).…”

“…From the remaining fractions the contents of the appropriate tubes were combined and deionized on small columns (15 cm. x 0-8cm.2) of Zeo-Karb 215 (H+) according to the method of Alfthan & Virtanen (1955). The amino acid eluates were evaporated to dryness on a steam bath, the residues dissolved in a little water and portions of these solutions checked for purity by paper chromatography in solvent system 2.…”
