Estudo de métodos de aumento de resolução de espectros de FTIR para análise de estruturas secundárias de proteínas

Abstract: FTIR. The FTIR spectroscopy has been used to quantify the secondary structures of proteins, using amide I band (1600 -1700 cm -1 ). The resolution enhancement methods have been used to resolve the individual components of this band that correlate to the secondary structure. In this paper we discuss the methods of derivative, Fourier deconvolution and fitting with simulated spectra. The results shows that they have serious problems and can be used only as a qualitative or semiquatitative method. Keywords: method… Show more

“…The signal at 1620 cm −1 (Fig. 1, curves b and c), as determined by the second derivatives, is assigned to the β sheet structures 20–23. These data indicate that the β sheet structure comes from the β zeins in samples CGM1 and CGM3.…”

“…The amide I signal of Z1 (α zeins, Fig. 1, curve a) is symmetric and the second derivative shows a strong signal at 1656 cm −1 , which is related to a high α helix content 20–23. The amide I spectra of CGM1 and CGM3 are not symmetrical and present a shoulder below 1656 cm −1 .…”

“…By calculating the Fourier Transform (FT) the interferogram can be written as a sum of sines and cosines (Tarumi et al, 2005) and in this case, happens to be called transmittance spectra, T (Forato;Filho;Colnago, 1997). Finally, the spectrum of transmittance, T, is converted to absorbance spectra, A, by co-logarithm of T (Suarez et al May 2011).…”
