Ethanolic Extract ofVitis thunbergiiExhibits Lipid Lowering Properties via Modulation of the AMPK-ACC Pathway in Hypercholesterolemic Rabbits

Abstract: Vitis thunbergii (VT) is a wild grape that has been shown to provide various cardioprotective effects. The present study was designed to examine whether a VT extract could reduce serum lipid levels and prevent atherogenesis in a hypercholesterolemic rabbit model. At the end of an 8-week study, our results showed that a VT extract supplement markedly suppressed the serum levels of cholesterol and low-density lipoprotein, reduced lipid accumulation in liver tissues, and limited aortic fatty streaks. Our findings… Show more

“…In total, 1 × DSW, and 2 × DSW significantly improved the TC level by 1.18-, and 1.21-fold, respectively. The results have been consistent with findings in other studies [28,29], and the results were similar to the effects in the MgCl 2 and lovastatin groups (Figure 1a).…”

“…Previously, Pan et al reported the methods for the quantification of atherosclerotic lesions in the aortic root [29]. To determine the effect of DSW on the fatty streak formation, six-week-old male New Zealand white rabbits were fed with the 0.5% cholesterol diet for eight weeks.…”

“…Composition analysis revealed that the dry root of V. thunbergii contained several kinds of polyphenols. Moreover, Pan et al [12] reported that ethanolic extract of V. thunbergii exhibited lipid lowering properties possibly by the activation of 5 -adenosine monophosphate-activated protein kinase (AMPK). Since AMPK is a critical regulator of glucose and lipid metabolism, our results and relevant findings suggest that the events of hypoglycemic and hypolipidemic effects might be pharmacological targets of polyphenolic compounds in ethanolic extract of V. thunbergii roots and the AMPK signaling pathway is a potential linking bridge.…”

“…The root of it has widely been used as folk medicine for treating fracture and injury, diarrhea, hepatitis and arthritis in Taiwan [8]. Recent studies have also indicated that it possesses a variety of efficacies such as antiplatelet [9], antimicrobial [10], antihypertensive [2], neuroprotective [11], and lipid-lowering [12] effect. These multiple health-promoting effects underscore the need to further investigate its action characteristics.…”
