Ethylparaben induces apoptotic cell death in human placenta BeWo cells via the Caspase-3 pathway

Abstract: Parabens are generally used as preservatives in foods, pharmaceuticals, and various other commercial products. Among them, ethylparaben has weaker estrogenic characteristics than endogenous estrogen. However, growing evidence indicates that ethylparaben has an adverse effect on various human tissues. Here, we investigated whether ethylparaben induces cell death by affecting cell viability, cell proliferation, cell cycle, and apoptosis using the human placenta cell line BeWo. Ethylparaben significantly decrease… Show more

“…Similar observations have been documented in other cell types, such as Vero and MCF-10A cells, 12,22 while propylparaben exposure has also been linked to reduced survival in human placental BeWo cells. 9 These findings underscore a complex relationship among cell type, exposure duration, and hydroxybenzoate concentration, resulting in varied levels of cell inhibition. The effects on cell survival are critical given the pivotal role of trophoblast cells in maintaining the placental barrier.…”

“…14 viability of human placental BeWo cells in a dose-dependent manner and induces apoptosis by enhancing caspase-3 activity. 9 Furthermore, it has been observed to impact the viability of mouse oviductal secretory epithelial cells, impeding cell cycle progression and reducing progesterone receptor levels in MOE cells. 15 Our research endeavors to provide a more comprehensive understanding of the effects of propylparaben on the trophoblast at the maternal−fetal interface.…”

“…To measure the mitochondrial activity of HFDP cells, cells were cultured in 96-well plates and treated with WEML. After 24 h, 20 μL MTS solution (Promega, Madison, WI, USA) was added and cells were incubated at 37°C for 1 h (Kim et al 2020 ). Absorbance was measured at 490 nm using a microplate reader (ABS Plus; Molecular Devices, San Jose, CA, USA).…”

“…The fraction of apoptotic cells was determined by flow cytometry. Apoptosis activity (%) was calculated by the percentage of apoptotic cells (annexin V-positive and PI-negative) (Bae et al 2020 ; Kim et al 2020 ).…”
