“…was confirmed by N-terminal and internal amino acid sequence analysis of polypeptides with an apparent molecular mass of 18.6 kDa from the purified complex. This size is consistent with a 14.5-kDa polypeptide that was identified immunochemically as COX II in cytochrome c oxidase isolated from C. reinhardtii (35).…”

“…To rigorously assay for transfer of proteins from the chloroplast to vacuoles, appearance of chloroplast-en-coded proteins in vacuolar granules was measured. Previous work (Hoober and Stegeman 1976, Rochaix et al 1989, Bennoun et al 1995 established that cycloheximide, at the concentrations used in our experiments, completely inhibited cytosolic protein synthesis in these cells and that synthesis of those proteins not affected by cycloheximide was completely inhibited by chloramphenicol, an inhibitor of chloroplast and mitochondrial protein synthesis. Thus, cbn1-113 arg2 cells were suspended in medium containing cycloheximide but lacking arginine and labeled with [ 14 C]acetate.…”

“…The difficulty of purifying mitochondria free of chloroplast contamination has limited biochemical research on Chlamydomonas mitochondria, and methods for purification of active mitochondria have been developed only recently (14,53). The 15.8-kb mitochondrial genome is linear and contains only a few genes: cob, cox1, five subunits of mitochondrial NADH dehydrogenase, the mitochondrial rRNAs (which are fragmented in the DNA sequence); three tRNAs, and an opening reading frame that resembles a reverse transcriptase (see GenBank Accession number U03843 for complete sequence and citations).…”
