Like all developmental processes, odontogenesis is highly complex and dynamically regulated, with hundreds of genes co‐expressed in reciprocal networks. Tooth agenesis (missing one or more/all teeth) is a common human craniofacial anomaly and may be caused by genetic variations and/or environmental factors. Variants in PAX9, MSX1, AXIN2, EDA, EDAR, and WNT10A genes are associated with tooth agenesis. Currently, variants in ATF1, DUSP10, CASC8, IRF6, KDF1, GREM2, LTBP3, and components and regulators of WNT signaling WNT10B, LRP6, DKK, and KREMEN1 are at the forefront of interest. Due to the interconnectedness of the signaling pathways of carcinogenesis and odontogenesis, tooth agenesis could be a suitable marker for early detection of cancer predisposition. Variants in genes associated with tooth agenesis could serve as prognostic or therapeutic targets in cancer. This review aims to summarize existing knowledge of development and clinical genetics of teeth. Concurrently, the review proposes possible approaches for future research in this area, with particular attention to roles in monitoring, early diagnosis and therapy of tumors associated with defective tooth development.

Objectives: The identification process is very important to determine the identity of the corpse. Identification through teeth is a primary identification method that is often used because teeth have a high degree of individuality. Identification through teeth can be analyzed through dental anomalies. Dental anomalies can assist in the identification process because of their uniqueness, stability, and high incidence in different populations. This paper aims to provide information on the importance of dental anomalies as a relevant and scientific means for identification purposes. Review: Dental anomalies can be defined as morphological abnormalities of the teeth that occur at the stage of growth and development of teeth. Dental anomalies are also inherited disorders that can occur as part of a syndrome. The wide variety of dental abnormalities in the form of size, number, morphology, structure or position of teeth in the oral cavity that can be possessed by each individual can be a characteristic of that individual. Conclusion: The use of dental anomalies in identification has been scientifically recognized with proper diagnosis and writing of dental anomalies in the medical record can be used for identification purposes in the forensic field.

Disturbances in histodifferentiation and morphodifferentiation may include genetic or environmental factors. These disturbances often lead to dental anomalies. This paper reviews the aetiology, clinical and radiographic appearance, diagnosis and treatment options for macrodontia and dens invaginatus respectively. A rare case is presented in which the two anomalies occurred directly next to each other in the anterior dentition of a young boy. The diagnostic process and management are reported with a detailed explanation of the use of modern diagnostic aids and equipment. The procedure of apexification is also described in detail as both anterior teeth were still immature when they turned necrotic. A 2-year follow up reported the successful treatment resolution of infection and associated symptoms, as well as restoration of function and aesthetics.
